Molecular Weight:
~100 kDa observed. The calculated molecular weight of this protein is 83 kDa; however, it has been observed at ~100 kDa in some cell lysates (Gruss, O. J. et al. (2002). Nat Cell Biol. 4(11);871-879.).
Immunogen:
Recombinant protein corresponding to human TPX2.
Background Information:
Targeting protein for Xklp2 (TPX2) is a microtubule formation and bundling protein that activates Ser/Thr kinase Aurora A during mitosis and targets it to the mitotic spindle. This effect on Aurora A, along with microtubule nucleation, is mediated by the N-terminus and C-terminus of TPX2, respectively. TPX2 is also required for normal assembly of microtubules during apoptosis.

Species Reactivity:
Human
Species Reactivity Note:
Demonstrated to react with Human.
Application Notes:
Immunoprecipitation Analysis: A representative lot from an independent laboratory immunoprecipitated TPX2 from Xenopus M phase extract (Gruss, O. J., et al. (2001). Cell. 104(1):83-93.). This publication used a purified form of this antibody when performing the IP. Cat. No. ABT177 is an unpurifed form of this antibody.
Immunocytochemistry Analysis: A representative lot from an independent laboratory detected TPX2 in a time-dependent assay using HeLa cells arrested in S phase (Gruss, O. J. et al. (2002). Nat Cell Biol. 4(11);871-879.). This publication used a purified form of this antibody when performing the ICC. Cat. No. ABT177 is an unpurifed form of this antibody.

Control:
HepG2 cell lysate
Quality Assurance:
Evaluated by Western Blot in HepG2 cell lysate.
Western Blot Analysis: A 1:1,000 dilution from a representative lot detected TPX2 in 10 µg of HepG2 cell lysate.
Purification Method:
Unpurified
Presentation:
Rabbit polyclonal serum with 0.05% sodium azide.
Storage Conditions:
Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
