Specificity:
This antibody recognizes FOXO1 phosphorylated at Thr24.
Molecular Weight:
~78 kDa observed.
Calculated molecular weight is 70 kDa. FOXO1 has been shown to migrate to ~78 kDa in western blots (Kamei, Y., et al. (2004). JBC. 279(39):41114-41123).
Epitope:
Phosphorylated Thr24
Immunogen:
KLH-conjugated linear peptide corresponding to human FOXO1 phosphorylated at Thr24.
Modifications:
Phosphorylation
Background Information:
Forkhead box O (FOXO) transcription factors play an important role in modulating metabolic functions and cell proliferation. Given the relatively high expression of FOXO1 in insulin-responsive tissues, this transcription factor is highly poised to regulate energy metabolism. When nutrient and insulin levels are low, FOXO1 promotes expression of gluconeogenic enzymes. Conversely, in the fed state, insulin levels rise and stimulate uptake of glucose primarily into skeletal muscle and other organs, including adipose tissue. Under certain pathophysiologic conditions, including insulin resistance, negative signaling to FOXO1 is compromised. Increased cell proliferation occurs when FOXO1 is inactivated when phosphorylated by Akt at Thr24.

Key Applications:
Western Blotting
UniProt Summary:
Function: Transcription factor which acts as a regulator of cell responses to oxidative stress. In the presence of KIRT1, mediates down-regulation of cyclin D1 and up-regulation of CDKN1B levels which are required for cell transition from proliferative growth to quiescence.
Subunit structure: Interacts with LRPPRC. Interacts with SIRT1 and this interaction requires the presence of KRIT1.
Subcellular location: Cytoplasm. Nucleus. Note: Shuttles between cytoplasm and nucleus.
Tissue specificity: Ubiquitous.
Induction: Expression is regulated by KRIT1.
Post-translational modification: Phosphorylated by AKT1; insulin-induced. IGF1 rapidly induces phosphorylation of Ser-256, Thr-24, and Ser-319. Phosphorylation of Ser-256 decreases DNA-binding activity and promotes the phosphorylation of Thr-24, and Ser-319, permitting phosphorylation of Ser-322 and Ser-325, probably by CK1, leading to nuclear exclusion and loss of function. Phosphorylation of Ser-329 is independent of IGF1 and lea |
