Product Overview:
Tissue inhibitor of metalloproteinases-1 (TIMP-1) has been demonstrated in numerous tissues and body fluids as one of the important regulatory factors in connective tissue metabolism. TIMP-1 is a glycoprotein (M.W. 32 kDa) that inhibits collagenases, gelatinases and stromelysin activities, and that is identical with collagenase inhibitor of b1-anticollagenase.
TIMP-1 may be quantitated using this sensitive immunoassay.
Test Principle:
The TIMP-1 immunoassay kit is a solid phase enzyme immunoassay (EIA) based on one-step sandwich immunoreaction. Upon addition of TIMP-1 containing specimen, the result is a TIMP-1 molecule being sandwiched between the solid phase and enzyme labeled antibodies. After removing unbound enzyme-labeled antibody, the well is then incubated with enzyme substrate and o-phenylene-diamine (OPD), resulting in the development of color. The activity of peroxidase enzyme is proportional to the amount of antigen, so that TIMP-1 concentration in specimens can be determined from a standard curve.
Application:
The TIMP-1 immunoassay kit is useful for the determination of TIMP-1 levels in fresh human serum and other body fluids. Antibodies in the kit are also reactive with bovine TIMP-1.
Contents of this kit are sufficient for assay of 96 samples, including standard curve. Testing of samples in duplicate or triplicate is strongly recommended. This kit is intended for research use only; not for diagnostic or therapeutic applications.

This gene belongs to the TIMP gene family. The proteins encoded by this gene family are natural inhibitors of the matrix metalloproteinases (MMPs), a group of peptidases involved in degradation of the extracellular matrix. In addition to its inhibitory role against most of the known MMPs, the encoded protein is able to promote cell proliferation in a wide range of cell types, and may also have an anti-apoptotic function. Transcription of this gene is highly inducible in response to many cytokines and hormones. In addition, the expression from some but not all inactive X chromosomes suggests that this gene inactivation is polymorphic in human females. This gene is located within intron 6 of the synapsin I gene and is transcribed in the opposite direction.
