Product Overview:
The TIMP-2 immunoassay kit utilizes antibodies immobilized on a bead matrix, in combination with enzyme-labeled antibodies, directed against different antigenic sites on the same TIMP-2 molecule. Upon addition of a TIMP-2-containing specimen, the result is a TIMP-2 molecule being sandwiched between the solid phase and enzyme labeled antibodies. After removing unbound enzyme-labeled antibody, the bead containing the sandwich is incubated with enzyme substrate and TMBZ, resulting in the development of color. The activity of peroxidase enzyme is proportional to the amount of antigen, TIMP-2, so that TIMP-2 concentration in specimens can be determined from a standard curve.
Application:
The TIMP-2 immunoassay kit is useful for the determination of TIMP-2 levels in fresh serum and conditioned medium of cells.
This system does not recognize TIMP-2 complexed with proMMP-2. While the immunoreactivities of TIMP-2 complexed with active MMP-1, MMP-2 or MMP-9 are increased by approximately 300%, 450%, and 300%, respectively, compared with that of the free form of TIMP-2, no change is observed when TIMP-2 is complexed with active MMP-3.
Contents of this kit are sufficient for assay of 100 samples, including standard curve. Testing of samples in duplicate or triplicate is strongly recommended. This kit is intended for research use only; not for diagnostic or therapeutic applications.

This gene is a member of the TIMP gene family. The proteins encoded by this gene family are natural inhibitors of the matrix metalloproteinases, a group of peptidases involved in degradation of the extracellular matrix. In addition to an inhibitory role against metalloproteinases, the encoded protein has a unique role among TIMP family members in its ability to directly suppress the proliferation of endothelial cells. As a result, the encoded protein may be critical to the maintenance of tissue homeostasis by suppressing the proliferation of quiescent tissues in response to angiogenic factors, and by inhibiting protease activity in tissues undergoing remodelling of the extracellular matrix.
