Background Information:
The Bmi-1 protein is a member of the polycomb group (PcG) proteins. Bmi-1 has been reported as an oncogene by regulating the cell cycle inhibitors p16 and p19. Bmi-1 knockout in mice results in defects in hematopoiesis, skeletal patterning, neurological functions, and development of the cerebellum. Bmi-1 is necessary for efficient self-renewing cell divisions of adult hematopoietic stem cells as well as adult peripheral and central nervous system neural stem cells. Bmi-1 may regulate the self-renewal of other types of somatic stem cells, and it is also thought to inhibit aging in neurons through the suppression of p53. Several lines of evidence implicate Bmi-1 in tumorigenesis. Differential expression of Bmi-1 in a mouse model of metastatic prostate cancer vs. wild-type suggests Bmi-1 expression levels may predict response to therapy and survival in multiple types of cancer.
