IgG
Background Information:
PI3 Kinase (Phosphatidylinositol 3-Kinase) is responsible for phosphorylation of the 3 position of the inositol ring of PI(4,5)P2, to generate PI(3,4,5)P3, a potent second messenger required for survival signaling, and insulin action. PI3 Kinase is a heterodimeric complex composed of an 85 kDa regulatory subunit and a 110 kDa catalytic subunit. Tyrosine phosphorylation of growth factor receptors creates docking sites for binding of p85 (through its SH2 domains) on the receptors; p85 brings with it p110, which is then proximal to its phospho-lipid substrate on the membrane. PI3 Kinase is also activated by Ras, and by the β:γ subunits of heterotrimeric G-proteins. PI3 Kinase is inhibitable by wortmannin, a useful tool for the study of the PI3 Kinase signaling pathway.

Species Reactivity:
Human
Species Reactivity Note:
Proven to react with human. Not believed to cross-react with mouse or rat. Reactivity with other species has not been confirmed.
Application Notes:
Immunoprecipitation:
A 1:40 dilution of a prevoius lot of this antibody immunoprecipitated PI3 Kinase, p110δ using protein G:agarose beads.
Immunfluorescence Staining:
A 1:250 dilution of a previous lot of this antibody was used for Immunfluorescence staining of paraformaldehyde fixed HeLa cells.

Control:
Jurkat cell lysate
Quality Assurance:
Routinely evaluated by Western Blot on Jurkat cell lysate.
Western Blot Analysis:
A 1:500 dilution of this lot detected endogenous PI3 Kinase, p110δ in lysate from Jurkat cells.
Purification Method:
Purified
Presentation:
Purified rabbit monoclonal IgG in buffer containing 60% storage buffer (50 mM Tris-Glycine (pH 7.4), 0.15 M NaCl, 0.01% sodium azide and 0.05% BSA) and 40% glycerol.
Storage Conditions:
Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20ºC. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20ºC may cause glycerol-containing solutions to become frozen during storage.
