Background Information:
Histones play a critical role in transcriptional regulation, cell cycle progression, and developmental events. Histone acetylation/deacetylation alters chromosome structure and affects transcription factor access to DNA. Histone Deacetylase 3 (HDAC3) belongs to the histone deacetylase/acuc/apha family. It has histone deacetylase activity and represses transcription when tethered to a promoter. It may participate in the regulation of transcription through its binding with the zinc-finger transcription factor YY1. This protein can also down-regulate p53 function and thus modulate cell growth and apoptosis. HDAC3 is regarded as a potential tumor suppressor gene that also plays critical roles in S phase progression and DNA damage control.

Species Reactivity:
Human
Mouse
Rat
Species Reactivity Note:
Demonstrated to react with human, mouse and rat HDAC3.
Application Notes:
Immunoprecipitation:
1:50 dilution from a previous lot was used for immunoprecipitation.
Control:
K562 whole cell lysate.
Quality Assurance:
Routinely evaluated by Western Blot on K562 whole cell lysate.
Western Blot Analysis:
A 1:10,000 dilution of this lot was used to detect Histone Deacetylase 3 in K562 whole cell lysate.
Purification Method:
Unpurified
Presentation:
Unpurified rabbit monoclonal IgG in buffer containing 50 mM Tris-Glycine (pH 7.4), 0.15 M NaCl, 40% Glycerol, 0.01% sodium azide and 0.05% BSA.
Storage Conditions:
Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
