Specificity:
This antibody recognizes AMPylated Ras-related C3 botulinum toxin substrate 1.
Molecular Weight:
~22 kDa observed. Other AMPylated proteins may appear at a different molecular weights.
Epitope:
AMPylated Threonine
Immunogen:
KLH-conjugated linear peptide corresponding to the Rac1 switch I region with AMPylated threonine-35 residue of AMPylated Ras-related C3 botulinum toxin substrate 1.
Background Information:
The PTM AMPylation, previously known as adenylylation, is the covalent addition of an Adenosine MonoPhosphate group to a hydroxyl side chain of threonine, tyrosine, and potentially serine residues. AMPylation of the threonine residue on such GTPases as; Rho, Rac and Cdc42 could interfere with the interaction of effectors downstream and shut down actin assembly in an infected cell. This antibody against AMPylated threonine has great potential to help in the identification of proteins containing this posttranslational modification.

Species Reactivity:
Human
Species Reactivity Note:
Demonstrated to react with Human. Broad species reactivity expected.
Application Notes:
Immunoprecipitation Analysis: A representative lot was used by an independent laboratory in IP. (Yi-Heng, H., et al. (2011). Journal of Biotechnology. 151:251–254.)
Control:
HeLa cell lysate incubated with 10 ng GST-VopS in the presence of 0.5 mM ATP
Quality Assurance:
Evaluated by Western Blot in HeLa cell lysate incubated with 10 ng GST-VopS in the presence of 0.5 mM ATP.
Western Blot Analysis: 1:2,000 dilution of this antibody detected AMPylated Threonine on 10 µg HeLa cell lysate incubated with 10 ng GST-VopS in the presence of 0.5 mM ATP. (Yi-Heng, H., et al. (2011). Journal of Biotechnology. 151:251–254.)
