Background Information:
Methylation of histones plays a critical role in regulating chromatin structure and transcription. The JMJD1 (Jumonji domain-containing protein 1) family, also known as JHDM2 (JmjC domain-containing histone demethylation protein 2) family, contains four members: hairless (HR), JMJD1A/JHDM2A, JMJD1B/JHDM2B, and JMJD1C/JHDM2C. JMJD1B has been shown to specifically demethylate mono-methyl and di-methyl histone H3 on lysine9. H3 Lys 9 methylation is a highly studied histone mark that represses gene expression and implicates JMJD1B as a key regulator of heterochromatin assembly. Deletions in the gene encoding JMJD1B are frequently associated with myeloid leukemias and myelodysplasias, suggesting JMJD1B also may act as a tumor suppressor.
