HAT Assay Reagent Kit
Description:
HAT Assay Reagent Kit
Trade Name:
Upstate (Millipore)
Qty/Pk:
1 kit
Product Overview:
Accessory reagent kit for the analysis of histone acetyltransferase activity.
Applications:
Accessory reagent kit for the analysis of histone acetyltransferase activity.
Key Applications:
Histone Acetyltransferase Assay (HAT)
Application Notes:
Nuclear Extract (source of HAT enzyme activity): Prepare nuclear extracts using a modified protocol of Dignam, et al., Nucleic Acids Res. 22: 1475, 1983 (recommended, but other methods may be used). Use 1- 20μl per assay point in the HAT Reaction Cocktail below. Optimal nuclear extract concentration will need to be determined empirically.

Entrez Gene Summary:
Histone acetylation, particularly of histone H4, plays an important role in replication-dependent chromatin assembly. This gene encodes a histone acetylase that contains A, B, and D motifs, present in many N-acetyltransferases, including those that acetylate substrates other than histones. Alternatively spliced transcript variants that encode different isoforms have been identified for this gene.

UniProt Summary:
FUNCTION: SwissProt: O14929 # May play a role in telomeric silencing. Acetylates soluble but not nucleosomal H4 at 'Lys-5' and 'Lys-12' and acetylates histone H2A at 'Lys-5'. HAT1 has intrinsic substrate specificity that modifies lysine in recognition sequence GXGKXG.
SIZE: 419 amino acids; 49513 Da
SUBUNIT: Heteromer of HAT1 and p46/HAT2 subunits.
SUBCELLULAR LOCATION: Nucleus. Cytoplasm. Note=Nuclear in S-phase cells and cytoplasmic.
SIMILARITY: SwissProt: O14929 ## Belongs to the HAT1 family.
