Background Information:
Histones are highly conserved proteins that serve as the structural scaffold for the organization of nuclear DNA into chromatin. Histone modifications regulate DNA transcription, repair, recombination, and replication. The most commonly studied modifications are acetylation, phosphorylation, methylation, and ubiquitination. These modifications can alter local chromatin architecture, or recruit trans-acting factors that recognize specific histone modifications (the "histone code" hypothesis). Global loss of acetylation at Lys16 is a common hallmark of human tumor cells, suggesting a role for acetylated Lys16 in preventing cell transformation. Moreover, Lys16 is the only histone mark that has been directly linked to chromatin refolding and maintainence of chromatin structure. The Sir3 protein helps form telomeric heterochromatin by interacting with hypoacetylated histone H4 Lys16 while the MYST-like acetyltransferase Sas2p is required for the acetylation H4-Lys16 in euchromatin.
