Background Information:
erbB3 (HER3, c-erbB3) is a receptor tyrosine kinase (RTK). erbB3 is one of four members of the EGFR/ErbB family of receptor tyrosine kinases. The ErbB family of receptors are a subfamily of four closely related receptor tyrosine kinases: EGFR (ErbB-1), HER2/c-neu (ErbB-2), Her 3 (ErbB-3) and Her 4 (ErbB-4). Though erbB3 is membrane bound and can bind its ligand, it does not have an active kinase domain so it can not transmit this signal without first forming a heterodimer with other EGFR family members. This heterodimerization then leads to proliferation and differentiation. erbB3 is also known to bind Phosphoinositide 3-kinase (PI3 Kinase, PI3K) though six tyrosine phosphorylation sites that PI3K can bind to, which results in the activation of Akt/PKB. erbB3 is known to be amplified in numerous cancers including breast and prostate.
