Background Information:
MAPKAP Kinases are Ser/Thr kinases activated by phosphorylation by the SAPK2/3, or p38 family of stress-activated kinases. MAPKAP Kinase 2 and 3 phosphorylate substrates with the sequence H-X-R-X-X-S where H is any hydrophobic residue, including CREB (Ser 133) and HSP27. MAPKAP kinases are similar to Rsk kinases, which are activated by mitogen-signaling pathways, rather than stress-signaling pathways. MAPKAP Kinase 2 is involved in many cellular processes that include inflammation, stress, and prolifertion.

Species Reactivity Note:
Human, Mouse, Rat. Other species have not been tested.
Application Notes:
Immunofluorescence Confocal Microscopy Analysis:
NIH/3T3 cells were fixed, permeabilized, and stained using Anti-phospho-MAPKAPK2 (Thr334), clone 9F10.2 (Red). Cells were co-stained for actin and nuclear staining with Phalloidin, AlexaFluor® 488 conjugate (Green) and DAPI (Blue), respectively
Control:
HeLa nuclear extract
Quality Assurance:
Routinely evaluated by Western Blot
Western Blot Analysis:
This lot detected MAPKAPK2 when phosphorylated on Thr334 at 1:1,000 dilution in PC12 lysates resolved via SDS-PAGE and transferred to PVDF (Immobilon-P).
