Specificity:
Recognizes ROKα/ROCK-II, Mr 150 kDa. Does not recognize ROKβ/ROCK-I. A non-specific band may also be detected, Mr 80 kDa.
Molecular Weight:
150 kDa
Immunogen:
6His-tagged fusion protein corresponding to residues 2-543 of rat ROKα/ROCK-II.
Clone:
A9W4
Background Information:
Rho-Associated Kinase (Rho-Kinase or ROK) has been classified into two isoforms: ROKα/ROCK-II and ROKβ/ROCK-I. These large cellular proteins (150-160 kDa), have been reported to be involved in the formation of stress fibers, focal adhesions and cardiac hypertonic responses. Current research suggests that ROK of smooth muscle may be involved in the contractile process via phosphorylation of MYPT1 and Myosin.

Species Reactivity:
Human
Mouse
Rat
Bovine
Species Reactivity Note:
Human and rat. Predicted to cross-react with mouse and bovine based on sequence homology.
Application Notes:
Immunoprecipitation:
4 μL of a previous lot immunoprecipitated ROKα/ROCK-II from 500 μg of PC-12 lysate.
Control:
RIPA lysates from PC-12 cells
Quality Assurance:
Routinely evaluated by Western BLot on RIPA lysates from PC-12 cells.
Western Blot Analysis: A 1:500-1:5000 dilution of this antibody detected ROKα/ROCK-II in RIPA lysates from PC-12 cells.
Presentation:
Cultured supernatant containing 0.05% sodium azide.
Storage Conditions:
Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
