TTK (also known as Mps1; monopolar spindles checkpoint kinase ) is a ~97 kDa cell cycle-regulated kinase and an essential component of the mitotic spindle checkpoint. TTK/Mps1 is shown to have maximal activity during M phase and minimal expression in anaphase where it becomes a target of anaphase promoting complex (APC). TTK/hMps1 is a Chk2 kinase where it binds and directly phosphorylates Chk2 kinase at threonine 68. TTK is phosphorylated on several serine and threonine sites including threonines 33 and 360, and serines 37, 362 and 363. Once phosphorylated, TTK/Mps1 is localized to kinetochores where it executes its checkpoint function and regulates multiple cell-cycle related processes, including cell cycle arrest.
