ELISA Amplification System
The ELISA Amplification System provides increased sensitivity (at least 10-fold) over traditional alkaline phosphatase-based ELISAs using pNPP. In the ELISA Amplification System, bound alkaline phosphatase acts directly on the substrate NADPH, whose product initiates a secondary cyclic enzyme reaction, resulting in a colored product (Figure 1). Each molecule of product from the first reaction takes part in many cycles of the second reaction; thus, the signal is amplified (Figure 2). The system produces a soluble colored end product and is therefore not suitable for immunoblotting or immunocytochemical applications. Streptavidin-alkaline phosphatase conjugate is supplied as a control. One system is sufficient for ten 96-well microtiter plates.
