PCR SuperMix High Fidelity
PCR SuperMix, High Fidelity, provides qualified reagents for high-fidelity amplification of nucleic acid templates by the polymerase chain reaction (PCR). You need only add template and primers, reducing set-up time by half. It is effective for either short or long targets. PCR SuperMix, High Fidelity, contains Mg++, dNTPs, and a mixture of recombinant Taq DNA polymerase and the proofreading DNA Polymerase from Pyrococus species GB-D, at concentrations sufficient to allow amplification during PCR. You will produce products that are mixed blunt and 3-A' ends; however, the majority of ends will have 3-A' overhangs.
Using PCR SuperMix, High Fidelity
PCR SuperMix, High Fidelity, is supplied at a 1.1X concentration to allow approximately 10% of the final reaction volume to be used for the addition of primer and template solutions. This ready-to-use reagent offers a 6-fold improvement in fidelity over Taq DNA Polymerase and is effective in amplification over a large range of target sizes. Targets beyond 12 kb in length may require optimization of PCR cycling conditions and success is heavily dependent on primer and template quality. Reagents sufficient for 100 amplification reactions of 50 µL each are provided.

PCR SuperMix High Fidelity Components: A 22 U/ml DNA polymerase mixture in 66 mM Tris-SO4 (pH 9.1 at 25°C), 19.8 mM (NH4)2SO4, 2.2 mM MgSO4, 220 µM dGTP, 220 µM dATP, 220 µM dTTP, 220 µM dCTP and stabilizers.
