This novel peptide sequence, RRRADDSDDDDD, is optimized for phosphorylation by casein kinase II (Km=19 µM). This peptide is not phosphorylated well by casein kinase I. The three N-terminal arginine residues allow it to be used in phosphocellulose kinase assays.
