Description pUC19 DNA/MspI (HpaII) Marker is recommended for sizing and approximate quantification of small linear double-stranded DNA fragments in agarose and non-denaturing polyacrylamide gels. pUC19 DNA is digested to completion with the appropriate Fermentas PureExtreme® restriction enzyme, then purified and dissolved in storage buffer. The DNA fragments contain blunt or sticky ends, depending on the restriction enzyme used for the markers preparation. pUC19 DNA/MspI (HpaII) Marker, ready-to-use, is premixed with 6X DNA Loading Dye for direct loading onto agarose and polyacrylamide gels. Quality Control Well-defined bands are formed during agarose gel electrophoresis. The absence of nucleases is confirmed by a direct nuclease activity assay. Storage and Loading Buffer (for ready-to-use markers) 10 mM Tris-HCl (pH 7.6), 10 mM EDTA, 0.005% bromophenol blue, 0.005% xylene cyanol FF and 10% glycerol. 6X DNA Loading Dye 10 mM Tris-HCl (pH 7.6), 0.03% bromophenol blue, 0.03% xylene cyanol FF, 60% glycerol and 60 mM EDTA.
