This lectin (MAL) is the leucoagglutinin or mitogenic isolectin form from Maackia seeds. This multimeric lectin dissociates into 38,000 and 40,000 molecular weight bands in SDS gels in the presence of reducing agents and is a single 75,000 dalton band without reducing agents. It has an isoelectric point about pH 4.6 and has been reported to have a molecular weight of about 130,000 by gel filtration. This lectin binds glycoconjugates having galactosyl (b-1,4) N-acetylglucosamine structures. Maackia amurensis lectin I seems to tolerate substitution of N-acetyllactosamine with sialic acid at the 3 position of galactose. However, MAL I does not appear to bind this structure when substitution with sialic acid is on the 6 position of galactose. |
